Sibylla Biotech is developing a new class of small-molecule protein degraders designed to unlock previously undruggable targets in oncology and beyond.
Its proprietary platform, PPI-FIT (Pharmacological Protein Inactivation by Folding Intermediates Targeting), uses atomistic protein-folding pathway simulations to identify transient folding-intermediate states of target proteins — structures that appear only during folding and are not accessible in the native, fully folded protein. This allows Sibylla to design small molecules, called Folding Interfering Degraders (FIDs), that lock these proteins into an intermediate state, triggering their elimination via the cell’s normal protein degradation pathways.
Sibylla’s lead programme targets Cyclin D1, a protein amplified and overexpressed in a range of cancers and long considered undruggable using conventional approaches. The company is also advancing a broader pipeline across oncology and other high-unmet-need therapeutic areas, since the PPI-FIT approach is in principle applicable to any target with a folding process.
